With my limited knowledge..few things I don't understand..
Mixer? Are you referring to studio/DJ kinda mixer, say with multiple Mic/line inputs, sound overlap, fade in/out..functions like that..or simply referring it as a graphic equalizer?
What kind of sub woofer you have, active or passive?...Do you have an active cross-over and an amp for the sub? (most of the active subs got both cross-over and amp build-in)
Would be the best if you can upload some pics of your gears, especially where the connectors are. I believe it is a lot easier for Mr. 7up to help you..
MACKIE DFX.12 12 channel integrated live sound mixer. (for karaoke)
ENERE e:XL-S8 subwoofer. at the back of the subwoofer. There are 2 round plug-in holes.
One in white colour with XOVER INPUT at the side.
One in red colour with SUB INPUT at the side.
My question is which hole should I plug in the cable from the mixer ?

You do have an active sub with 100 watts amp build-in but no XLR connections. However, on the upper right side of the mixer where it says main out, there is XLR thingy. Maybe you should consider XLR conversion as MR. 7 UP suggested. I am a straight line Hi Fi dude, seeing so many controls cause me headache... Should check with 7UP.. He da best..hehe.

I see some RCA jacks near there too, but they are not main outs (CD/Line in & Tape out). Means, without XLR conversion, your gear and sub won't "talk" to each other..
If you are using AV amp, just plug into the sub out of AV amp, that is the easiest..

Your sub also has hi level inputs, (not the option I like the most) but lots connect this way. Reason why is the sub is seeing the same signal as main speakers..

Diagram one is most likely will work..2, and 3...Hmm...sub has build-in cross over. #4 not sure if he's got pre-out main-in...doubtful..
Lazy bastard, hehe...Just kidding..hope you don't confuse him... Not the best option, but it will work.. Lack of info for other possible connections anyway.
